Ly6G (Lymphocyte antigen 6 complex locus G6D) is a component of the myeloid differentiation antigen Gr-1, together with Ly6C. The Ly6G protein has a molecular weight of approximately 21-25 kDa and is expressed by myeloid derived cells in the bone marrow. Ly6G is a good marker for detection of peripheral neutrophils, monocytes and granulocytes.

Gr-1, and more specifically RB6-8C5, the monoclonal antibody that recognizes Gr-1, has been a very important tool for immunologists investigating neutrophil function in murine models. RB6-8C5 binds to 2 members of the Ly6 family of leukocyte-expressed markers, Ly6C and Ly6G.

Ly6G was identified as a novel member of the Ly6 family in 1993 [58, 68]. Ly6G is a small protein of 25 kD that is tethered to the cell membrane via a GPI linker . Unlike the Gr‐1 antigen, which encompasses epitopes from Ly6G and Ly6C, expression of Ly6G is markedly restricted. 2017-06-26 · Thus, Ly6C hi cells are often termed “inflammatory monocytes.” In contrast, the nonclassical monocytes (CCR2 – CX3CR1 ++ Ly6C lo) do not express CCR2, but do express the highest levels of CX3CR1 . These monocytes patrol blood vessels and also accumulate at inflammatory sites, where they are thought to give rise to M2 macrophages . Ly6C is a monocyte/macrophage and endothelial cell differentiation antigen regulated by interferon gamma and is thought to play a role in the development and maturation of lymphocytes.

Markers such as CD11b, CD11c, F4/80, Gr-1, Ly6C, and Ly6G have long been used to identify various splenic cell myeloid populations. Flow cytometry and fluorescence-activated cell sorting (FACS) analysis demonstrated that Ly6G/Ly6C markers are superior to Gr-1 for identifying splenic neutrophils, eosinophils, and subsets of monocytes/macrophages.

The rat monoclonal antibody RB6-8C5 detects Ly6G component of Gr-1 antigen, a commonly used surface marker of neutrophils. Figure B. As we know, CD11b + Ly6g + Ly6c int cells are recognized as PMN-MDSCs (immature neutrophils) in mouse. However, ly6g is expressed on neutrophils as well. The question is how can we distinguish the

The RB6-8C5 monoclonal antibody reacts strongly with mouse Ly6G and weakly with mouse Ly6C previously referred to as GR-1.

Immunogen corresponding to tissue, cells or virus. 2021-02-12 Ly6G ligation on murine neutrophils inhibits neutrophil recruitment, thus providing the first evidence of a function for the Ly6G molecule. Ly6G is seen primarily on neutrophils, also in a subgroup of eosinophils, differentiating pre-monocytes and plasmacytoid dendritic cells. 2017-10-31 Currently, there is no standardized panel for immunophenotyping myeloid cells in mouse spleen using flow cytometry.
